Cervomed Inc. reported a mean reduction of 3.1 pg/mL in plasma neurofilament light chain levels at week 12 among patients with the non-fluent variant of primary progressive aphasia, according to clinical data presented at the 15th International Conference on Frontotemporal Dementia in Philadelphia.
Clinical Trial Results in Philadelphia
The Nasdaq-listed clinical-stage biopharmaceutical company presented open-label data from its ongoing phase 2a study evaluating neflamapimod in patients with the non-fluent variant of primary progressive aphasia. The trial enrolled 25 participants to assess the oral small-molecule therapy’s impact on key neurological damage biomarkers. According to the presentation, the overall average reduction of plasma neurofilament light chain—known as NfL—occurred predominantly between the sixth and twelfth weeks of dosing.
In a pre-specified exploratory subgroup analysis of 13 patients whose baseline NfL levels sat at or above the median, the 12-week drop was more pronounced, showing an average decrease of approximately 5.7 pg/mL. Meanwhile, glial fibrillary acidic protein levels—serving as a secondary biomarker—remained stable throughout the initial 12-week monitoring window.
Safety Profile and Tolerability
Investigators observed that neflamapimod maintained a generally favorable safety and tolerability profile, with no novel safety signals emerging during the evaluation period. One trial participant discontinued treatment after experiencing an elevation in liver enzymes. That participant’s laboratory values returned to baseline following cessation of the drug, without concurrent symptoms or increases in bilirubin levels.
The experimental treatment targets p38 alpha MAP kinase, an enzyme involved in neural inflammation and neurodegeneration within the brain. Cervomed operates from its headquarters in Boston, Massachusetts, focusing development efforts on age-related neurological conditions.
Timeline of Upcoming Data Releases
The company outlined its upcoming milestones for the phase 2a program during the Philadelphia conference. Cervomed intends to release 24-week biomarker findings at the upcoming Clinical Trials on Alzheimer’s Disease conference in November. Following those intermediate data points, the organization expects to announce topline clinical results for the study in the first quarter of 2027.
