Ultimately, Henderson decided to accept an offer from Ohio State University and play college football for the Ohio State Buckeyes. In his freshman year, he developed into a regular player at the running back position under head coach Ryan Day. In his first game as a starter, he was able to run the ball for 277 yards, setting a new university record.[5] Overall, he ran the ball for 1,248 yards and 19 touchdowns in his freshman year.[6] After being slowed down by injuries in his sophomore year, he was able to build on the good performances of his first season in his last two years.[7] Overall, he ran the ball for 3,761 yards and 42 touchdowns, making him the fifth-leading running back in school history behind Archie Griffin (5,589 yards), JK Dobbins (4,459 yards), Ezekiel Elliott (3,961) and Eddie George (3,768).[6] He also scored 6 receiving touchdowns, making him the third most with a total of 48 touchdowns after Pete Johnson and Keith Byars. Due to his good performances, he was elected to the First-Team All-Big Ten in 2023, to the Second-Team All-Big Ten in 2021 and to the Third-Team All-Big Ten in 2024.[2] He was also very successful with his team: in 2025 they won the College Football Playoff National Championship Game and thus won the college football championship. They also won the Rose Bowl in 2021. After the 2024 season, Henderson declared that he would enter the NFL draft.

In the 2025 NFL Draft, Henderson was selected in the second round, 38th overall, by the New England Patriots.[8] At the beginning of the season he was listed as the second running back behind Rhamondre Stevenson.[9] He made his NFL debut on Game Day 1 of the 2025 season in the 13:20 defeat against the Las Vegas Raiders, in which he ran the ball five times for 27 yards. On Game Day 4, Henderson finally scored his first touchdown in the NFL in a 42:13 win against the Carolina Panthers.[10] On the 9th matchday he was a starter for his team for the first time, and on the 10th matchday he even scored two touchdowns in the 28:23 win against the Tampa Bay Buccaneers. Just a week later, he even recorded three touchdowns, two rushing and one receiving, in a 27:14 win against the New York Jets.[11] He then became… AFC Offensive Rookie of the Month November appointed.[12]
