Berrettini won his opening match at the 1000 tournament in Canada 6-4, 6-3 against Gregoire Barrere, Musetti had previously won 6-4, 6-1 against Yoshihito Nishioka.
Berrettini is allowed to test another Italian in round 2, namely compatriot Jannik Sinner.
Jiri Lehecka, Francisco Cerundolo and Alejandro Davidovich Fokina were also among the early winners on Day 1 in Toronto. Still in action on European nights are Gael Monfils and Tommy Paul, among others.
Toronto without Thiem, but with Zverev
The top seeded players only get involved from Tuesday, with the top eight players having a bye in Round 1. Kitzbühel finalist Dominic Thiem will not start in Toronto, Alexander Zverev will start with Washington finalist Tallon Griekspoor.
Top seeded is Wimbledon champion Carlos Alcaraz ahead of Daniil Medvedev. Novak Djokovic is currently pausing.
